2024.05.19 00:16 OmegaThree3 SAM-e Review… Helps!

My only major mast cell activation symptom is swollen nasal turbinates, which I can only really control with wearing a breathe right nasal strip and nasalcrom and DAO before meals. If I am wearing the nasal strip, I have no symptoms, it just looks goofy.
I have tried things like all the natural herbal stuff (quercetin, luteolin, nettle), ketotifen, and then methylation things like methyl folate, TMG, and betaine HCl nothing really has any effect whatsoever.
Recently, I tried 400 mg of SAM-e and it made my nose open up! But I also slept really shitty. I then purchased a 200 mg version since I can’t cut the pills it seems as they are enteric coated. And it helps open the knows as well. I can go without the nasal strip during the day without any discomfort, which is new. However, it’s still fucking up my sleep.
Insomnia is one of my main symptoms due to tickborne infections specifically Bartonella, so I don’t know what’s better having an open nasal passage or the bad sleep but I’m glad I at least found something that works.
I am not sure how I am going to proceed, but just wanted to share my reaction. Let me know your Intel on SAM-e and what you think?
Did I miss anything I should try?
I’m not sure the mechanism, and although traditionally, I am sensitive to things that boost Neuro transmitters. I think this is more of a downstream effect, but maybe the increase in serotonin is potentially keeping me up at night.

Chronic UTIs for 6 months
Going to try and keep this short.
I’ve been experiencing recurrent/chronic UTIs since November. At this time I started seeing my boyfriend, so I have been sexually active.
I’ve been prone to UTIs since I first became sexually active in high school, I’m 25 now. I take every precaution. I drink water. I pee right after sex. I take D Mannose. I’ve tried everything.
I’ve officially been on antibiotics 7 separate times since November. I just finished a long dose of Macrobid, 2 daily for 10 days and my symptoms have remained.
I had a urinalysis and it came back positive for white blood cells, but no growth.
I’m at my wits end. It’s beginning to affect me mentally because I’m in constant pain and fear that when I have sex I’ll make everything worse.
I’m at the point where I’m considering going to the hospital because i’m not sure if it’s an infection that’s resistant to antibiotics, or if it would be a waste of time to go if I’m getting negative urinalyses.
I’m looking for any sort of advice. I work a full time in-office job, and it’s starting to affect my work as well.

So, I have done a lot of research on it and ordered one with my FSA card as they had a sale on one of their kits.
Getting genetic testing through my doctor would take so much effort to convince them that I looked at outsourcing it myself. I found Sequencing.com and so far, I like everything I see. Tbh the website is super helpful even if you don't get a screening from them.
I have a slew of issues - some diagnosed and some not.
I'm auDHD (late diagnosed) and I have Psuedotumor Cerebri (IIH). I've had it for the past 8 years with no relief. I'm 27 years old and at the same weight I was when I graduated high school. My BMI is 26.whatever and I'm pre-IIH weight so I "should" be in remission.
Through subs like this one, I learned about Mast Cell Activation Syndrome and holy shit is that my entire life. But it's a blood test for genetic markers to confirm or even a bone marrow test for the markers.
I went to my (usually great) neuro for my 2 month follow-up to get my lidocaine/steroid shots in my neck and lower back. I have two slightly shifted discs that pinch nerves. I was having the symptoms of a CSF leak (I've had one before and know how it feels) and he gaslit me with someone else's medical history (saying I had it checked out before and my ENT said I was fine) except I haven't been to my ENT since I had mono two years ago and that was 2 years AFTER my blood patch that had me in the hospital 3 days.
I'm rambling. I'm really excited about this test and all the stuff it could tell me. I got the ehlers-danlos syndrome kit for $400 which gives me a 3 week turn around on my test too.
I really enjoy that the founder is a doctor who had a similar issue as me: unknown but chronic illness undiagnosed for years.

I was looking up how to buy a Brawl Pass so I could give one to my friend as a graduation gift, I’ve been playing the game for a month or so and this is the first time I’ve made a purchase here. I just assumed the Brawl Pass would appear in an ‘inventory’ or something under my SuperCell ID so I could donate it to my friend.
I DIDN’T realize that the game would automatically activate the pass and add all the currency to my account. The (one-time) refund policy is a 5,000 gem subtraction to your balance (not worth it) and support seems extremely stubborn on maintaining this policy, so I guess I’m stuck with a bunch of items I don’t want for an accidental purchase. I really wish that there was a differently licit in place, is it really that difficult to subtract the accidentally purchased items from the account instead?

• Health Rising reported some time ago becoming obsessed with the energy problems found in ME/CFS. Vishnu Shankar, a PhD. Stanford student, engineered a study that assessed energy production in ME/CFS patients’ immune cells. The study recently appeared in preprint form and includes some new findings – so it’s onto round 2 of this fascinating study.
• Shankar focused on immune cells because they provide a good test case for assessing energy production. It turns out that protecting the body from pathogens turns is quite an energy-intensive project. Even when it’s not fighting off an infection, the immune system uses about 15-20% of our energy.
• The study assessed both mitochondrial activity and oxidative stress in immune cells (PBMCs) in ME/CFS patients, long-COVID patients, and healthy controls.
• The immune cells in the ME/CFS/long-COVID patients weren’t pumping out as much energy as the healthy controls’ cells – suggesting they were damaged and possibly exhausted.
• Because damaged mitochondria can become free radical-producing machines, they took a look at oxidative stress (free radical) levels. (Just as a damaged automobile engine produces more exhaust than a well-functioning one, damaged mitochondria spew out more toxins; i.e. free radicals.)
• Immune cells have to switch their after-burners on to get the energy to go after pathogens. Producing that energy comes at a cost, though, in the form of increased levels of reactive oxygen species oxidative stress (free radicals). (Note the key word – reactive oxygen species – these are unbalanced oxygen molecules which try to achieve balance by ripping electrons from other molecules; hence the word “reactive”.)
• Dramatically higher levels of reactive oxygen species were found in both the ME/CFS and long-COVID patients’ immune cells compared to the healthy controls. (ME/CFS patients had the highest levels). A closer look revealed, however, that the reactive oxygen species were almost wholly centered in the immune cells from female ME/CFS patients.
• Both male ME/CFS and long-COVID patients did, however, exhibit elevated glutathione levels – indicating that their immune cells were also dealing with increased oxidative stress levels – which have triggered the production of the master antioxidant in our cells – glutathione.
• Altogether, multiple pathways that deal with oxidative stress (glutathione, superoxide dismutase, lipid oxidative damage, etc.) appear to have been overwhelmed in the immune cells in both ME/CFS and long-COVID patients. In all cases, the ME/CFS patients were worse off than the long-COVID patients – leading the authors to suggest that long COVID was an intermediate condition between health and ME/CFS.
• Increased levels of mitochondrial calcium – which drive the production of reactive oxygen species (ROS) in the mitochondria – provided another potential explanation for the increased levels of oxidative stress. That was a very interesting finding given that in 2021 Wirth and Scheibenbogen proposed that increased mitochondrial levels of calcium were a core factor in ME/CFS.
• Once again, damage to the lipids that protect our cells and the organelles in our cells popped out during a metabolomic analysis (of T-cells). Since reactive oxygen species target lipids this finding make sense and it underscored what big deal lipid issues have become in ME/CFS studies over the past few years.
• Once again, men and women had different findings. Oxidative stress was worse in women while lipid damage was more extensive in men. Plus, the study suggested that when confronted with high oxidative stress levels, women’s T-cells go on a hyper proliferation binge – potentially sucking more energy from the rest of the body.
• Trying to reduce the oxidative stress present, they used antioxidants like NAC (increase glutathione), metformin (increase SOD2 expression), and liproxstatin-1 (reduce lipid peroxide levels) in cell cultures and found that NAC and metformin was able to reduce immune activity to some extent. That finding suggested that the right antioxidants might be able to tame the immune activity and improve energy levels.
• All in all, the authors proposed that long periods of elevated reactive oxygen species damage the mitochondria, producing a long-term problem of energy depletion. Because the immune cells already use up so much energy in the body, and then struggle to produce energy in ME/CFS and long COVID, the authors proposed that immune problems in these diseases produce an energy sink that draws energy from other areas of the body.

These notes come from watching every ppshow this week and taking my favorite bits of tinfoil from the community into one fairly simple look into the story dfv is telling us without getting too deep on possible tinfoil, it’s just laid out plain and simple here.
Tldr; this is a very long post. I think it starts with dfv’s final yolo update (fine I’ll do it myself) and how he’s been feeling for the first 30 minutes of the memes and then in the last half it switches to, fine I’ll do it again and then full of straight confirmation foil that we’ve been right about the bear trap, bbby, Teddy, and baby all along. Then he says we’re all good, be zen and says goodbye for now.

So I’m on the King of Puppets, and I can pretty consistently get through the first phase no problem, some runs I make it with 2 pulse cells remaining, one time I made it hitless to Romeo, but regardless I’m working on learning this boss and beating him legit. I’m also playing around with using my Specter and Grindstone. Here’s where tips come in:
With the wishetone, is this an item that I should activate before crossing the fog wall, is this an item that’s activated mid fight, is it always for the specter or do some stones apply to P. A lot of the translations or descriptions I feel are so vague I’m left questioning exactly what it does i.e. P-Organs/Wishstones

Let me preface by stating that I love this sub! I find Taylor's relationship with Travis to be completely fabricated and dishonest, and I'll be lucky if they last less than a year. It all sounds fake, PR influenced, and just for the benefit of putting Travis in front of cameras and for Taylor to show this newly "sporty/different" side of her after her breakup with Joe. Hopefully the mods here can see my post history (I was decently active on swiftlyneutral and I can see through the flaws and problems that Taylor showcases in her personal life, while still enjoying her music as a casual listener).
Anyways to my point: my girlfriend is obsessed with Taylor. Almost to a mood altering parasocial levels of Stanning her. If Taylor is rightfully criticized for X (take your pick: carbon emissions, relationship w/ Matty Healy, Brazil's show, Fake PR relationship) she will be in a bad mood for a few days until it dies down, because Taylor's problems always get swept under the rug. Recently we were discussing TTPD and she mentioned how "The Alchemy" and "So high school" are so so so good that she believes it's proof that Taylor's next album, which she heavily believes will be about Travis Kelce, will be a fantastic, amazingly written, chart topping new Era album that will be a highlight of Taylor's career.
I'm sorry but in a whole DOUBLE album that you originally alluded to being the "takedown" of Joe, the guy who you were with for 6 years, you have: 1. A song where you mention his depression and mental health being reasons for why you couldn't stay with him 2. The majority of the album being about Matty, who you fantasized about so badly and who you wanted to be with SO BAD that you basically cucked your current PR boyfriend and 3. Two songs about the aforementioned PR BF, one being decent (the alchemy) which wholly focuses on the only personality trait that Travis exudes (loud football jock) and the other being an extremely badly written song about you guessed it, high school (the mental age that both Travis and Taylor are stuck in).
I'm sorry but "I know Aristotle" and "touch me while your bros play Grand Theft Auto" MAKE ZERO SENSE and if any other artist had said that, it would put their pen game in question. how does my girlfriend ( who I love with all my heart) see this and says "huh, fantastic writing, it's so in depth and just shows how amazing and great their relationship is". You could only play two player GTA until GTA 4 came out with online play (in late 2008). Why would they be playing GTA instead of idk, Madden? A game you can actually take turns playing? To me, it just reaffirms that Travis has literally no personality. He is the walking embodiment of a guy who only likes football, getting drunk, and plays video games ( not sure if it's confirmed, but I'm pretty sure he still hasn't beaten GTA San Andreas, the plane missions are too hard for him IYKYK). Him having no personality also reaffirms how fake and PR this relationship is!!
TLDR; GF thinks two bad songs on Taylor's new Album = They are in it for the long run and next album will be a classic. Lyrics show otherwise. Their relationship Is a PR stunt, and I would like to sit in as a ghost on one of their conversations to see how many brain cells Travis can make me lose in 20 minutes.
Would like to add: this relationship is clearly just to keep attention away from Taylor and Matty Healy, right? Because her being with him would ruin her brand and public figure image that her PR team has been carefully manufacturing ever since 2016, right?
Final note: is the eras tour just her version (no pun) of when guy's get their heart broken and go to the gym to get fit and focus on themselves? Or to a lesser extent when you have a bad day and you go for a long run? Feels like it

I have a Google Sheet which contains various worksheets:

The worksheets contain a variable amount of data, but typically have between 1-6 columns, where A is the Code, B is the Modifier, C is the Description, D is the Abbreviated Description, and E are any applicable Notes. Because this data is updated very frequently, I want to use Apps Script to automate several processes to streamline maintenance of this workbook:
When a code is entered in a cell in column A, execute the following steps:
  1. Evaluate column A for duplicate entries; if the code already exists, reject the newly entered (duplicate) code
  2. Sort the worksheet by column A in alphabetical order
  3. Capitalize any alphabetical characters in the code string; for example, if I enter 3074f, change it to 3074F
In the past, it seemed to work to some degree, but now nothing is happening as expected.
Gemini was able to get the automation to work without requiring the use of a trigger (somehow), but in its troubleshooting suggestions, it said I need to create one. I have done so, and it still will not work--I keep getting an error when I debug the function.

I'm graduating with a PhD in chemistry in July and have been searching for jobs in biotech/pharma. The positions are usually scientist I-III with roles in canceimmune research.
While my degree is in chemistry, our lab does little traditional chemistry (e.g. chemical synthesis), and instead focuses on areas such as immunotherapy, biophysical, and analytical microscopy. I'm not sure if the fact my PhD is in chemistry rather than immunology or something similar is hurting my chances or if my experience isn't competitive enough, my resume doesn't convey the relevant experience well, or if I'm not even making it past the ATS or recruiter.
I've applied to ~75 jobs since the beginning of April and have only have 1 real interview. I use a hybrid resume/CV (I read that's best for industry research position. I also try to tailor my resume for each application (putting most relevant experience first, using matching keywords and skills), but most of the positions are very similar and my resume doesn't change much.

This morning my Golden and I were at the large dog park throwing the ball. She’ll play with other dogs and chase each other but she does her own thing too. Not an aggressive bone in her body and she’s not annoying to other dogs either because she’s never too interested or focused on them. A woman, I didn’t recognize, came into the gates with a pitbull mix. She’s got her headphones in and glued to her phone. I’m already grabbing my things to leave. Her dog starts trying to aggressively play with mine and barking and nipping; basically harassment as my dog is avoiding it trying to ignore it. So now I’m on alert. The pit has my dog’s ball in its mouth. I ask the owner to get the ball from her dog because I’m leaving. She pulls a treat out of her bag to distract her dog who instantly drops the ball for the treat. My dog then goes for his dropped ball and that’s when all hell broke loose.
I’ve seen quite a few altercations between dogs at the park but when it’s a pitbull it’s an entirely different terrifying situation. Immediate switch to a death machine. Growled a deep strong growl for about 1 second before it leaped on my dog attacking. I jumped to grab my dog and get her away from this pit. The woman gets hers still trying to attack mine. I rush my dog out of there and what does the woman do? She LET GO of her dog to continue to enjoy the park and she went back to her cell phone w her ear pods in!! What the actual fuck!? I wish I yelled at her I wish I made a scene to get her to leave but I was too shaken up.
Some other info: I’m inexperienced so when I decided to get my first dog last year I decided to buy a well bred dog of the friendliest breed. My main concern was ever having to deal with aggression as I am not a professional trainer. So, I got a Golden Retriever! Funny, how many dog owners do not prioritize that when getting a dog. I still took her to training school, hired a private trainer and heavily socialized her.
I know dog parks aren’t typically recommended but I have a 1yr old Golden Retriever who needs tons of activity and exercise. I take her to this one dog park because it’s a massive enclosed field and typically the dog community there are responsible owners and well behaved dogs. And lots of regulars so you know the majority of the people and dogs.

Introduction
Unlike many other places in the world, the USA uses a completely different set of LTE bands for cellular communications, additionally, American carriers impose device restrictions, which may adversely impact uses of feature phones, as they often are heavily locked down to the carriers from which they came originally, or have to be imported. This guide simply outlines my experiences dealing with the three major carriers (AT&T, T-Mobile, and Verizon) and general assumptions about regional carriers (such as U.S. Cellular and Viaero Wireless).
AT&T
AT&T is potentially the worst provider for feature phone users, given that AT&T enforces a whitelist of devices which may be used on their network (PDF: read the whitelist on KONAT.xyz, mirroring the original on att.com), which impairs the use of feature phones as only a handful of models (mainly AT&T/Cricket branded [in-house] models, US-market Nokias, and rugged/work-oriented devices from manufacturers such as Kyocera). From experience, AT&T's enforcement of the whitelist is generally extremely strict, to the extent that your account may be suspended for using non-whitelisted devices.
Even if a device works on AT&T's network and is whitelisted, it may not have additional features (such as WiFi calling) functioning on the network. Additionally, carrier variants of devices (i.e. the variant sold by T-Mobile) may not be whitelisted. Below is a non-exhaustive list of MNOs/MVNOs which do and do not enforce the whitelist; this list may become outdated at any time.
AT&T uses LTE bands 2, 4, 5, 12, 14, 17, 30, 46, and 66. In my experience, bands 12, 17 and 2 are the most useful to have in urban areas. Band 14 is used for the First Responder Network Authority (commonly referred to as FirstNet). I would not advise using a device without complete band compatibility outside of urban areas.
ENFORCES WHITELIST: AT&T post-paid, AT&T pre-paid, Cricket Wireless (the prior three are all owned by AT&T). The vast majority of MVNOs can also be assumed to be enforcing the whitelist.
DOES NOT ENFORCE WHITELIST: H2O Wireless, FreedomPop, RedPocket GSMA
T-Mobile
In general, T-Mobile can be argued to be the best provider for feature phone users, as it does not enforce a whitelist, and generally allows any device with the LTE bands used in a particular area to function on the network and utilize VoLTE, WiFi calling, or other carrier functions. To further simplify this, this means T-Mobile will allow the use of imported devices which have the band(s) required to function in a particular area, as well as devices from other carriers, in addition to US-market unlocked devices and devices sold specifically for T-Mobile's network.
T-Mobile also operates an EDGE/2G network and small slivers of HSPA/3G. The EDGE network is moribund at best (and hasn't been expanded in well over a decade, nor does it appear to be a going concern maintenance-wise), and the remaining slivers of HSPA are officially meant to be offline. T-Mobile's legacy networks should not be depended upon for reliable communication.
T-Mobile operates on the following network bands relevant to feature phone users: EDGE/2G: 1900 MHz (PCS) LTE: 2, 4, 5 (only utilized in Myrtle Beach, South Carolina), 12, 66, and 71.
From experience, devices lacking any band other than B2 will work in urban areas, but B12 is generally needed for indoors signal. B71 is needed for good coverage in rural areas. As a resource for figuring out which bands are in use in particular areas, take a look at the band rollout map.
For reference, MVNOs which operate on T-Mobile include Mint Mobile, Ultra Mobile, MetroPCS (the prior three all are owned by T-Mobile), Tello, and US Mobile.
Verizon Wireless
I would personally rank Verizon as second-best for feature phone users, as Verizon generally allows any device with LTE band 13 to utilize their network. Verizon does restrict new activations, however, this can be bypassed by activating a new SIM card in a cell phone that is known to work with Verizon (such as an Apple iPhone or similar known-working device) and moving the SIM card to the B13-having feature phone (or other non-accepted B13-having) device after confirming that the activation went through successfully.
Devices without B13 may work in urban areas, but may not have voice or SMS functionality (limited to data only), thus, it is a necessity to use a device with B13. Additionally, some MVNOs on Verizon's network (the only one I know of currently is TracFone and associated brands [Simple Mobile, Total Wireless/by Verizon, Straight Talk, et. al.) may bind the SIM card to the device it was activated in, and in this case will likely suspend accounts for moving the SIM, even into another 100% functional device, thus making it impossible to use devices that require the above activation method.
Verizon uses the following LTE bands: 2, 4, 5, 13, 46, 48 and 66
MVNOs that operate on Verizon's network include TracFone et. al., Visible Wireless (the prior two are owned by Verizon), US Mobile, and Xfinity Mobile.

So let's say theoretically the FBI is right and a pride parade is attacked either by a foreign or domestic group like Hamas or a cell of Nazis, and the police pull a Uvalde and don't intervine or actively keep assistance away from the attack, what are the potential "legal" consequences of this, if there are any at all? And if theres somehow no legal consequences, can be speculated that the backlash could be very violent and severe?

The fundamental purpose of eating is to replenish the chemical elements composing the cells and tissues in our body. Replenishment is one of the basic laws of Nature in regard to organic chemistry, and our physical body is a laboratory functioning under organic chemistry principles.

The food that we eat should nourish these cells and tissues. Unfortunately, because Nature gave to man a body so elastic, so far as taking punishment is concerned, that it can survive for years on food which is destructive to the body, but appealing to his appetite and palate, man has indulged these appetites until the race is perceptibly degenerating physically.

The body is the vehicle of the mind, and the mind is the vehicle of the intellect. The intellect is that part of the mind that we use in observing and reasoning.

If the body is permitted to degenerate, then the intellect cannot be expected to function or develop constructively, as the spiritual and mental faculties in man grow and expand in direct relation to the improvement, regeneration and purity of the physical body.

We look on sickness and disease as something mysterious and dreadful and we blame germs and bacteria.

As a matter of fact germs and bacteria are the scavengers of Nature and are everywhere. When we breathe, we inhale millions of these little natural scavengers and it is their function to keep the debris in our body neutralized and to stir it up so that it can be eliminated from the body. It is our job, however, to keep our body in such condition that this elimination can be completed to perfection.

Due to the excessive quantity of inorganic food that we eat, food in which the life principle has been destroyed by cooking, canning, and other processes, this debris, or end-product of the digestion of this food, in the body, accumulates faster than we and these natural scavengers can remove it. The result is that germs and bacteria find a feeding ground within us in which to propagate. In this process of their propagation the sewage of their colonies is added to the debris and the result s what we call sickness or disease.

Whenever germs and bacteria enter a body which is thoroughly clean and healthy, within and without, they find no feeding ground of waste or morbid matter therein on which to colonize and therefore pass out of the system in the natural course of events.

Likewise, when all debris and accumulated morbid matter is removed from a sick or diseased body, then only is the first step established toward a recovery to a normal chemical balance.

How does this debris and morbid matter get into the body? In two distinct ways.

First, through the food which is eaten to excess in inorganic form (cooked, etc.) which can neither be assimilated by the body for constructive purposes, nor be properly eliminated; and also by such unnatural elements as serums, vaccines, injections, etc. These cause deposits which the body is unable to throw off in the course of its normal functions, if the eliminative channels are in any way impaired.

Second, through the cells and tissues of the body which in the course of our activities are constantly used up and remain there as dead matter after furnishing physical and mental energy. These used-up cells should leave the body as soon as possible after they have served their function, but they remain in the system for unnecessarily long periods of time due to faulty elimination.

We can give here only briefly a few of the interesting reasons why nourishing the body properly is of such vital importance. A brief outline of the process that causes the greatest accumulation of debris in the body is necessary so that we may be guided intelligently in the selection and combination of our foods.

Let us take, for example, an individual, 40 years of age. Undoubtedly like the majority of his fellow men he has formed the habit of eating at least three meals a day. This will average more than 1,000 meals a year, or in excess of 40,000 meals during his lifetime of 40 years. It is very important that we bear this in mind.

We will also assume that, like most people, nearly all the food he has eaten has been cooked, canned or otherwise processed, and rarely, if ever, does he eat either a sufficient quantity of raw food, or a complete meal of nothing but raw vegetables and fruits. The result therefore is that more than 40,000 meals, composed mostly of dead food (or inorganic chemical elements) have passed through his system during that time.

It is impossible to regenerate organic cells in a human body with inorganic (or dead) matter. We find that while the 40,000 meals did serve the purpose of sustaining life, hardly any nourishment in organic form was eaten to regenerate the cells and tissues of his body or supply the chemical elements composing these.

As a matter of fact it is appalling to realize that the limit of tolerance of such a condition is usually reached by the time we are 40 or 50 years of age. The age at which mature judgment and experience have been gained, a period in life when we should know what life has in store for us, the very prime of life, but the age at which most men and women find themselves with a neglected body which is degenerating rapidly and is no longer an efficient vehicle in which to put that knowledge and experience into practical use, speedily heading for premature senility and the discard.

We know that the body requires fiber. The error in judgment is in the interpretation of what function the fiber is intended to perform. To be of any value whatever, the fiber in our food must be composed of the raw cellulose, or fiber, of raw vegetables and fruits eaten as nearly as possible in their natural ripe state.

When so eaten, after proper mastication, the digestive processes extract as many as possible of the chemical elements contained in the fibers. The remaining fiber goes through the intestines becoming, figuratively speaking, highly magnetized by means of the muscular kneading of their peristaltic action. Thus they draw into the intestines from every part of the body the used-up cells and tissues, eliminating these, as well as the debris resulting from normal digestion and the end-products thereof, through the colon, thereby acting both as an intestinal broom and as a vacuum cleaner.

When food has been cooked or processed in any manner or form, however, the fiber or cellulose is converted into an inorganic substance in which every vestige of life has been destroyed. The fiber being lifeless therefore can no longer work as a broom or as a vacuum cleaner, but operates instead like a mop (usually a slimy one) pushing matter along without any cleansing effect. Due to lack of magnetic attraction, which cannot be generated in dead fiber, it has no ability whatsoever to draw into the intestines used up cells and tissues from other parts of the body, nor any other toxic matter which may have accumulated therein.

Consider these two pictures. On the one hand an abundance of raw fibers passing through the digestive and eliminative tracts, acting as a cleansing broom and as a vacuum cleaner after every meal, three times a day, leaving not only a clean intestinal tract each time but also removing from the system other accumulated waste matter. On the other hand visualize the fiber of cooked, devitalized food (nearly always in excessive quantities) passing through the intestines and eliminative organs three times a day - 40,000 times or more in 40 years-every time leaving a coating. 40,000 coatings, be they ever so thin, are bound to leave their mark.

Man is the only member of the animal kingdom who, notwithstanding his supposedly higher intelligence, indulges his appetites at the expense of his body, and pampers these deliberately, without using common sense or good judgment, listening to the silky voice of deception telling him that food has nothing to do with the condition of his body.

With such a lack of knowledge it is not surprising that just when we have reached the age when our knowledge and experience are of value to us, for us to use to advantage, and we are really anxious to begin to LIVE, we find ourselves handicapped with a physical body which is ready for the discard, if not for the grave.

The problem therefore is to know how to change our eating habits to regenerate our body, without too much discomfort, and reactions too strong for the comfort of the mind or the vicissitudes of our daily life, and what will enable us to achieve eventually a healthy body free from fatigue, sickness and disease.

Chronic spinal cord injury (SCI) is difficult to cure, even by several approaches effective at the acute or subacute phase. We focused on skeletal muscle atrophy as a detrimental factor in chronic SCI and explored drugs that protect against muscle atrophy and activate secretion of axonal growth factors from skeletal muscle. We found that acteoside induced the secretion of axonal growth factors from skeletal muscle cells and proliferation of these cells. Intramuscular injection of acteoside in mice with chronic SCI recovered skeletal muscle weight reduction and motor function impairment. We also identified pyruvate kinase isoform M2 (PKM2) as a secreted factor from skeletal muscle cells, stimulated by acteoside. Extracellular PKM2 enhanced proliferation of skeletal muscle cells and axonal growth in cultured neurons. Further, we showed that PKM2 might cross the blood–brain barrier. These results indicate that effects of acteoside on chronic SCI might be mediated by PKM2 secretion from skeletal muscles. This study proposes that the candidate drug acteoside and a new myokine, PKM2, could be used for the treatment of chronic SCI.

Two major issues in chronic SCI are the untreatable axonal disruption and the severe atrophy of skeletal muscle; our study showed that both may be improved by acteoside, via PKM2 secretion. In this study, we found that acteoside injection into skeletal muscle at the chronic phase of SCI recovers locomotor dysfunction and skeletal muscle atrophy in mice, despite the fact that our contusion model was quite severe; less than grade 2 at 30 days post-injury. Our results indicate that PKM2 is a new myokine that activates skeletal muscle and neurons and that its secretion is enhanced by acteoside stimulation.

Further, we found that PKM2 might cross the BBB and BSCB despite its large molecular weight. After exercise, transcription of several myokines is increased in the brain. However, there is no clear evidence on whether myokines transfer to the brain. PKM2 may be a new type of myokine that reaches the brain and potentially the spinal cord. The advantages and significance of PKM2 penetration in the central nervous system should be clarified in future. The mechanism by which PMK2 crosses the BBB and BSCB might be similar to that in the case of IGF-1 (MW: 8.5 kDa), which crosses the BBB by transcytosis.

In cancer cells, acteoside directly binds to and inhibits protein kinase C. Moreover, binding to and inhibition of caspase-3 of acteoside were found in neurons. In inflammatory cells stimulated with lipopolysaccharide, acteoside promotes nuclear factor κB inhibition. Inhibition of calcium influx by acteoside has also been reported.

In conclusion, we found that acteoside improves skeletal muscle atrophy and locomotor dysfunction caused by chronic SCI, suggesting that it might be a promising therapeutic drug candidate for SCI. We also demonstrated that acteoside promotes the secretion of PKM2 from skeletal muscle cells, and that extracellular PKM2 induces axonal growth in cortical neurons and increases proliferation of skeletal muscle cells. A strategy involving skeletal muscle-mediated therapy is a novel approach for chronic SCI, which might be effective in combination with other treatments.

In addition, they found a new myokine called PKM2, or pyruvate kinase isoform M2, which promotes proliferation of skeletal muscle cells and the growth of axons in the brain. It crosses the blood-brain-barrier, potentially in a similar way to IGF-1, or insulin-like growth factor 1. The acteoside from Cistanche stimulates the release of PKM2. This is a really cool new mechanism for Cistanche that I was unaware of. After working on a bunch of different extraction methods, we found that supercritical CO2 was the best way to concentrate the highest amount of acteoside. The more we learn about Cistanche, the cooler of a supplement it becomes!

Chronic spinal cord injury (SCI) is difficult to cure, even by several approaches effective at the acute or subacute phase. We focused on skeletal muscle atrophy as a detrimental factor in chronic SCI and explored drugs that protect against muscle atrophy and activate secretion of axonal growth factors from skeletal muscle. We found that acteoside induced the secretion of axonal growth factors from skeletal muscle cells and proliferation of these cells. Intramuscular injection of acteoside in mice with chronic SCI recovered skeletal muscle weight reduction and motor function impairment. We also identified pyruvate kinase isoform M2 (PKM2) as a secreted factor from skeletal muscle cells, stimulated by acteoside. Extracellular PKM2 enhanced proliferation of skeletal muscle cells and axonal growth in cultured neurons. Further, we showed that PKM2 might cross the blood–brain barrier. These results indicate that effects of acteoside on chronic SCI might be mediated by PKM2 secretion from skeletal muscles. This study proposes that the candidate drug acteoside and a new myokine, PKM2, could be used for the treatment of chronic SCI.

Two major issues in chronic SCI are the untreatable axonal disruption and the severe atrophy of skeletal muscle; our study showed that both may be improved by acteoside, via PKM2 secretion. When we worked on our supercritical CO2 Cistanche, we found that extraction type was concentrating acteoside to the highest amount. This mechanism points to the idea that stacking our supercritical CO2 Cistanche with some other products that increase nerve growth factor, like Erinamax and Tiger Milk, could be a super effective stack!
