So I think clearly there was strong evidence that there was a significant event that happened in that laboratory in September. It's now been declassified, you can read it. I wanted to reach out to the community and ask for some suggestions. I have a sample which I IP'ed. We figured since the concentration was so slow and having difficulties getting a good purity ratio, we decided to amplify the sequence. Now it is in the post amplification process (primers attached still etc). I am wondering what everyone's favorite DNA purification kit is? I am having a hard time finding a very good DNA purification kit. I have looked into the AMPURE XP bead but it is way too expensive.... (plate + solution = ~2K.
Any suggestions are greatly appreciated

For lithium batteries or lithium battery packs, a total of 7 items of tests T1 → T5, T6, and T7 are required. However, for lithium battery cells, T1 → T5, T6 and T8 tests are required. Among them, T6, T7, and T8 tests need to use separate samples. And T1 → T5 test sequentially tests the same sample.

Comparison of UN 38.3 & International Standards

Compared with other international lithium battery standards, the temperature cycling conditions specified in the UN38.3 standard are more demanding and longer. Other international standard test items generally use separate samples for testing. However, the T1 → T5 test in the UN 38.3 standard tests the same sample in sequence. The previous test may have a negative impact on the next test, resulting in failure to pass the test. If the lithium battery submitted for inspection is tested for T1 → T5 in the UN 38.3 standard, there will be one item unqualified. Enterprises need to re-test UN3813 standard after process improvement, which will greatly extend the testing cycle.
It can be known from Table 1 that compared with the international standard IEC 62133, the high and low temperature exposure time of the temperature cycling item in the UN 38.3 standard is up to 6 hours, and the low temperature test temperature is -40 ± 2 ° C. I have a fusion biopsy scheduled for Friday. I've been reflecting on how I got here. Something like this:
2015 - Diagnosed with Type 2 Diabetes and sleep apnea.
2016 - Hypogonadism. Started TRT. PSA spiked. 1 of 12 samples mildly interesting. Started watchful waiting. Prostate measured 75 CC.
2017 - Excessive red blood cell production. Stop TRT.
2017 - 2022 - Focus on managing T2D and mental health issues.
2022 - Occasional ED. Treated with PDE5 inhibitors.
2023 - 6/23 Notice mild curve in erection. Curve not extreme, no painful erections.
2024 - 2/24 Best metabolic panel of my adult life - and a) wbc out of range on the high side and b) PSA spiked. Was 1.8 in 8/23, up to 8.4 in 2/24.
2024 - 3/24 Experiencing tiny urine leakage. No trouble using the bathroom otherwise.
2024 - 4/24 Prostate MRI. Results:
2024 - 5/24 - Erection curve more noticable.
My thoughts: There always could be PC lurking around. My younger brother had RALP 10 years ago, when he was 48. He did not have BPH. That and co-morbidities are a real distraction. It is difficult to suss out multiple causes and effects.

Was this potential suicide letter in Kris’s car coerced?
Was it her willingly writing it?
You don’t know.
As we have discussed in previous posts, nothing about the Kristin Snyder missing person case makes any sense whatsoever, and the purported “suicide note” found in her vehicle is certainly no exception.
Before we start analyzing the “suicide note,” here are a few factors to keep in mind:
• We know through information from multiple sources that Kristin was a prolific journal writer and letter writer, so we have a plethora of writing samples to compare with this alleged “suicide note.”
• We refer to “the writer” in our discussion of the “suicide note” below. The reason for this phrasing is because the actual writer of this note is unknown. Did Kristin herself write it, either as a explanation for killing herself, or for the purpose of faking her own death? Did someone else write it to make her disappearance appear to be a suicide? Was part of the text written by Kristin and added to by another party? Was the entire note faked? Was the note written by Kristin, but under duress/coercion as Heidi pondered?
• See notes under each section below regarding clear discrepancies between Kristin’s baseline writing style based on the hundreds of writing samples we have obtained from multiple sources through varying times throughout her life.

Similarly, wouldn’t she document the date of the most significant letter of her life, as she did routinely with her letters? In fact, she often even included the specific time (for instance, 7:15 p.m.) that the letter or journal entry was written.
• In addition to a salutation and date on other writing samples, Kris also typically indented her paragraphs and she also usually wrote on each line of the paper in her letters and journal entries, unlike this “suicide note,” which does neither.
• Related to the numerous writing samples we have acquired though multiple sources, Kris also primarily wrote in cursive in both her letters and in her journal. This “suicide note” is an odd hybrid of cursive and print.
• Why would anyone start a suicide note with “I attended a course…”? Clearly, the writer of this note is directing the reader to correlate ESP with the disappearance, but it seems like a very odd place for anyone to start a suicide note. Also, Kris attended two courses, not “a course”; a fact that Kris would have clearly known.
• “aka Nexium” is another oddity. Kris did not take any NXIVM classes, not even one, despite the extensive recent propaganda linking her to NXIVM. Why? Because NXIVM did not even exist at the time of Kristin’s disappearance; it was still in the planning stages. The writer had obviously heard about these plans as evidenced by the phonetic spelling. Again, it is obvious the writer of the note is clearly directing the reader’s attention to ESP/NXIVM – but if Kris were distraught enough to write a suicide note (and as functionally incapacitated as reported by her partner), why/how would she focus on minutiae like this?
• Speaking of minutiae, it gets even more obvious in the next words: “based out of Anchorage, AK + Albany, NY.” First of all, WHO CARES where ESP was based? That is in no way pertinent to the reasoning, and apparently is another clear attempt by the writer to direct the reader toward ESP/NXIVM. Secondly, this information is actually wrong. ESP wasn’t “based out of Anchorage, AK” – they held classes in Anchorage in a rented hotel space. The home base was in NY. Furthermore, Kris knew very well that this information was wrong, having recently visited their NY headquarters herself weeks before her disappearance!
•The words “based out of” (city, state) are odd as well. None of Kristin’s other writing samples did this. Nowhere does she mention elsewhere that anything is “based out of” anywhere in any of her copious writing samples we have obtained.
• Furthermore, why would the note say “Anchorage, AK” anyway? Presumably, Alaska law enforcement would be able to deduce that Anchorage is in Alaska without this unnecessary clarification.
“I was brainwashed + my emotional center of the brain was killed/turned off. I still have feeling in my external skin but my internal organs are rotting.”
• If Kris was brainwashed, she wouldn’t know (at least at the time) that she had been brainwashed. Again, this seems to be yet another clear attempt by the writer to direct the reader to look at ESP.
• Furthermore, if Kris finally did realize that she had been brainwashed, why would she then kill herself?
• The writer switches “my” and “the” in a sentence – something Kris never did, even once, in the hundreds of pages of writing we have obtained. The sentence should read “the emotional center of my brain,” not “my emotional center of the brain.”
• Another oddity is in the redundancy of “external skin.” Again, this sort of mistake does not appear to be Kris’s style, based on other writing samples. She had a Master of Science (M.S.) in Biology and she worked as an environmental consultant to the National Guard. She was a precise, clear, scientific, and articulate writer.
• This passage clearly implies that Kris was suffering from Cotard’s syndrome; per WebMD: “People with Cotard’s syndrome (also called walking corpse syndrome or Cotard’s delusion) believe that parts of their body are missing, or that they are dying, dead, or don’t exist.” We have talked to multiple people who Kris had visited in her January 2003 trip immediately prior to her February 2003 disappearance, and nobody reported any observations of any mental health issues, suicidal ideation, depression, psychosis, nor delusions of any sort. All of the people who discussed Kris’s reported mental health decline stated that they had not personally witnessed any symptoms, but rather, they were told of a rapid decline following Kris’s disappearance.
• If Kris thought she was already dead, why would she kill herself?
“Please contact my parents Bob + Jonnie Snyder at (number redacted) in Dillon, SC if you find me or this note.”
• Why would she specify to contact her parents, who lived out-of-state? Why not her partner? Why, in fact, is Heidi, the love of her life and civil union partner not mentioned AT ALL in the entire note?
• The inclusion of Kris’s parents as the sole contacts listed in the note contradicts a specific story told at the time of the disappearance alleging that Kris had uncovered memories of abuse during the class and that these purported memories were the reason/a factor in her alleged suicide. But: if that story was true, why would she include her father in the note? It should be noted that there is no evidence whatsoever that Kris was abused. As with the alleged rapid mental health decline, people who reported that story were not told of the purported abuse by Kris themselves, but rather, they were told of the purported abuse allegations after her disappearance. In fact, we even have been given a copy of a text message exchange in which the person who spread this abuse claims refers to it as “the lie.” This is yet another example of the myriad of inconsistencies and contradictions that plague Kris’s case.
• Why mention “Dillon, SC”? There is already a phone number given, so the city/state is irrelevant, and also, it is not her typical style. Again, it seems like someone with a quirky tic to mention a city and state wrote this.
• “if you find me or this note” is similarly nonsensical. If someone found her but NOT the note, they wouldn’t see the note, would they? Again, this oddity of wording is inconsistent with Kris’s typically precise style.
“I am sorry, life, I didn’t know I was already dead. May we persist into the future. KRISTN (sic) SNYDER”
• Again, if she thought she was already dead, why would she need to kill herself?
• Why is she addressing “life”?
• “May we persist into the future” is interesting. “Persist into the future” is a phrase used in ecology, which could potentially mean a couple things: a). Kristin wrote this herself; b). Kristin wrote this phrase elsewhere and someone traced/copied it onto the “suicide note”; or c). the writer had seen a document that referred to this phrase and used it.
• WHO LEAVES A LETTER OUT OF THEIR OWN NAME???? The second “I” is missing in “KRISTN.” Furthermore, as mentioned earlier, Kris predominantly wrote in cursive and she typically signed her name in cursive as well. Why, in the most important document of her life, would she BLOCK PRINT her name, and even more bizarre, why would she leave a letter out of her own name? The writer appears to drop letters and cram letters together, but there is no evidence from other writings that Kris did these things.
“No need to search for my body”
• Why was this written on the BACK of the page on the “suicide note”? And why was the note left inside of a notebook to begin with?
• Kris was a member of the Anchorage Nordic Ski Patrol, and therefore, she was involved in search and rescue. Therefore, she would already know that THEY WOULD SEARCH FOR HER ANYWAY. Also, more importantly, why would she intentionally hide her own body and therefore put her colleagues/friends on the search and rescue team through the extensive trouble and potential dangers of conducting the search for her?
• Why write “my body” on the back of the page but write “me” on the front of the page of the note? That is yet another incongruity.
• Why the emphasis on not looking for a body? The writer clearly has a very specific reason to mention this; there is a reason the writer does not want the body found. It is very rare for a person to want to hide his/her own body, and even more rare to be able to successfully do so.

2024.05.13 21:52 BroasisMusic Best practices for running a hardware synth through an MPC to my audio interface?

Hey all. I've got an MOTU M4 interface and a MPC Key 61, which is connected to the MOTU's rear input 3 and 4 via a pair of balanced TRS. I also have a Juno-X on the way. I don't want to use the two front inputs on my MOTU for either the Juno or the MPC, as I use them for easy guitar and microphone access. I could get an external mixer or a signal switcher, but would like to avoid if possible. I also don't want to use the Juno-X as my computers 'sound card', because then I'd lose the output to my monitors from my interface.
Is there any drawback - other than not having individual input channels in my DAW - to routing the balanced XLR output of the Juno-X to the two XLR inputs on the MPC, and then passing-through and monitoring/recording the signal via my MOTU? I'm assuming here my signal chain would stay balanced?
I'm hesitant to go this way, as I don't want anything in the MPC or the signal chain to color the sound from the Juno before it hits the interface. I'd like it passed through as cleanly as possible (I believe since it's Zencore powered, it's 24bit/48khz). How can I ensure any pre-amps or processing is disabled in the MPC if I go this route, and would this be a good enough way of getting the audio from my Juno to my interface in a simple and transparent manner? Or will there always be a downside, whether it's coloring the sound or some sort of sample rate conversion, or something I'm not thinking about?
I don't really use the DAW or sequencing function of the MPC Key, so I'm not interested in recording the Juno sounds into the MPC itself, just using it to monitor and pass along some preferably untouched audio from a hardware synth in this instance, so that I can record it via Ableton on my PC.

I’ve been craving a good enemies to lovers roleplay for awhile now and what’s a better enemies to lovers roleplay than something revolving around the mafia?
Plot idea #1: Your character has recently taken over the family business. My character is a college student on a study abroad trip that happens to see something they weren’t supposed to. Y/C’s men that were handling the deal take her back to the main ‘headquarters’ and keep her locked up, leaving Y/C to deal with her.
Plot idea #2: Your character is in a small gang that is tasked with kidnapping the naive daughter of a mob boss that stole money from another. Basic idea, can be fleshed out more together.
Plot idea #3: your character would be a police officer that’s gone undercover with some small local gangs to bust a couple of drug dealers and whatnot. Having done such a great job there, he is offered a bigger mission and if he takes it, when he returns to his actual station, he will get a bigger raise, new car, K-9 unit, some sort of special thing that he’s been working towards. This bigger mission would be infiltrating a bigger mafia family that seems to be running all these smaller ones. Thanks to some forged documents, lies spread around, and some of the actual dirty work he has done over the last six months, the head of the family invites him to a nightclub to chat. The chat leads to the offering of a position within the family, a personal bodyguard for the mob bosses daughter after the situation that happened with her last one. It’s his first break through and they’re welcoming him with open arms. Does he accept?

For the end of video demo Andrew says he recorded the Plonk onto the 4ms Stereo Triggered Sampler, loaded them onto his computer, sorted and refined them, then sequenced them in a DAW of his choice.
I would like to be able to do all of this in my Eurorack, preferably with one, if not two, module(s).
I was looking into using the Nerdseq with another sample recorder such as the Disting EX, but unfortunately I can't easily pan these samples left and right while pitch shifting as far as I know, ala Aphex Twin.
I've also though of getting a Polyend tracker for this, but I really would like to keep everything contained within my Eurorack case if possible.
The purpose of this will be to create complex drum sequences. What do you all suggest? Is there a module that possibly does all of this?

Hey All!
I'm working on a rather complex problem and have been for a considerable amount of time. A year more or less.
The data that I'm working with comes from a rather simple simulator that generates pairs of points on a cylinders wall (excluding the top and bottom). The pair of points defined by 2 3D locations that make up what we call a line-of-response (LOR) in the land of medical imaging. These lines intersect a very small volume (dependant of the particle that is placed in the PET scanner). Now there are algorithms out there that can find the location of these lines but I'm trying to build a neural network that can do this. The current start of the art method is using PEPT-ML (unsupervised machine learning). It does this by separating the total number of LORs into chunks of some size we can call N. It then computes the smallest connecting line. If this line is less than some user defined constant, we can call this MD (maximum distance), the centroid of this connecting line is then saved. These centroids are then passed into HDBSCAN which then computes clusters with the help of another parameter called TF (true_fraction) which is the ratio of inliers to outliers. You can then easily remove the noise that HDBSCAN identified and compute the mean of the remaining points in the cluster to find the location.
What I'm trying to do is remove the hassle of having to deal with and optimise all these hyperparameters. I've decided to create a neural network that can find the location within a sample. So I've tried many approaches such as a FFN, transformer encoder and lastly is the recurrent networks. The current network architecture is displayed below and currently provided the best minimum L2 loss . The network hyperparameters are input_dim = 6 (The 2 3D interaction locations), output_dim = 3 (the output location), num_layers = 4 (any larger doesn't work), hidden_dim = 256 (again any larger doesn't work).

The batch size being used is 1024 if that's relevant? Larger batch sizes tend to train a lot slower and with the amount of training data (1_000_000) and validation data (100_000) it already takes some time. I'd prefer if it doesn't take any longer to train.
I have uploaded the training and validation data to onedrive if anyone want's to have a look. Here is the link. Anyone should be able to get the data if they so wish. You should also find the script that is used for training.
The preprocessing of the data was performed by scaling the (x1, y1, z1, x2, y2, z2) between -1 and 1. This was applied to all pairs of interactions/LORs. It should be noted that The (N, M, 6) array was converted to a (2*N*M, 3) array first and then the scaling was applied and afterwards was converted back to a (N, M, 6) array. N denotes the amount of different samples of LORs, M denotes the number of sequences. This was done so that the targets can be scaled as well. Also this allows for unscaling of predictions during testing.

The loss looks as follows. Weird spikes. No idea what causes that. Also no idea why the validation loss is lower than the training loss. I don't think that's an issue?

The issue with this approach is that although the validation loss is considerably small it is still not good enough. A good way of thinking about this loss is that 0.1 loss is more or less equal to 10k mm error. 0.01 loss = 1k mm error. 0.001 loss = 100 mm error. 1e-4 loss = 10 mm error and so on. I think I need a loss of 10^(-8) or something.

Each of the scatter points are from the neural network being used to output a location from the sequence of LORs. It's really odd that the Z dimension is perfectly fine and more than acceptable but X and Y are well not great. I know there's a symmetry in X and Y but would that cause an issue?
