2024.05.16 22:24 PetVitalityApp Veterinarian: How I approach the allergic dog in my practice

Please note: I've mentioned some products and brands here just because I use them regularly in my work. Nothing has been received for their references in this article.
This is a simplified step-by-step guide to help dog owners understand the steps towards allergy diagnosis and management. Unless the logic behind the steps are communicated properly by the vet, I understand that the process may appear slow, frustrating and not always clear. It is important to keep a good dialogue with the vet throughout the process and not to expect a quick-fix after the first consultation.
Also note that each case is handled differently based on many factors such as the owner's financial limitations, country-specific norms and the veterinarians habits and experiences. Please note that this is just an example on how I myself approach allergic dogs. Other veterinarians may have completely different routines that works for them.
Common presentation:
Allergies typically begin to develop in dogs between the ages of 1 and 3 years old and usually present with one or more of the following symptoms:
There are plenty of differential diagnoses for each of these symptoms. For the sake of simplicity we must assume other conditions have already been ruled out, otherwise I would probably have to write a small book.
1: Anti-Parasitic treatment
Even if I strongly suspect allergies, I usually always start out an allergy consultations by ruling out parasitic infestations. A skin scraping can be performed and investigated under the microscope, but a negative skin scrape does not rule out parasites; it only rules out parasites at that small patch of skin where the sample was taken. It's a simple and cheap treatment, so if the issue happens to be parasitic it will save the owner a lot of time and money. If the symptoms are severe, I provide anti-itching drugs for a few days while the trial anti-parasitic treatment has time to work.
(Hypothyroidism can also cause allergy-like symptoms and should be ruled out (blood test) before initiating a full allergy investigation.)
2: Hypoallergenic diet trial:
If the symptoms are still present after anti-parasitic treatment course, I start the dog on a prescription Allergy diet for 8 weeks. We use Royal Canin Anallergenic since it's the most extensively hydrolyzed diet available. Hydrolyzed diets contain proteins that have been chemically 'chopped' into tiny subunits, so small that the pet's immune system cannot recognize them as allergens. Since they are not recognized by the immune system they will elicit no allergic reactions. During the food trial it is important that the dog ONLY eats the prescribed diet and avoids everything else, including treats and human foods. If the dog is severely affected I will provide anti-itching medicines for a few weeks at the start of the food trial, but it is important that the medicines do not mask the effect of the food change for us to know what actually worked.
If the symptoms disappear after the 8 week allergy trial diet, then great, we may conclude that food is the cause of the pet's allergies. I will then usually do elimination diets to figure out what types of food the dog actually tolerates. Some dogs may have to remain on prescription allergy diets for the rest of their lives. Some vets perform a food challenge test (Back to the diet they reacted on previously) after symptoms have cleared to check for recurrence as a way to confirm their diagnosis.
FOOD ALLERGY BLOOD TEST: These tests are somewhat controversial as they are NOT tools for diagnosis or identifying specific food allergens. However, they can be useful during a food trial in determining what protein sources the dog is less likely to have an allergic reaction to.
3. Environmental allergy screening + trial treatment.
If the allergy symptoms are still present after 8 weeks of prescription allergy diet, the cause of the allergy is more likely to be environmental (or both). At this point I usually recommend doing a blood test screening or panel against specific environmental allergens, such as pollen or mites. I always offer to do the blood test during step 2, but many clients prefer to go stepwise.
While waiting for the blood results I usually initiate treatment with Cytopoint injections. Depending on the severity I might add Apoquel short term for relief as it is very quick-acting and effective drug. For very severe cases corticosteroids might be required. I prefer Cytopoint for long term use over Apoquel and (especially) glucocorticoids due to having a more specific mechanism of action, while the alternatives are more broad-acting immuno-suppressive drugs. However, each patient responds differently - Finding the ideal treatment is usually a case of trial and error and there is no one-size-fits-all solution. Sometimes a combination is required.
NOTE: Neither Cytopoint or Apoquel cure allergies. They provide relief to the pet by blocking some of the symptom's pathways. However they are still very important medicines for improving the quality of lives for thousands of pets with allergies.
\ Cytopoint: Injections lasting 4-8 weeks.*
\ Apoquel: Daily tablet or chewable.*
4. Immunotherapy:
Immunotherapy is the closest we currently get to 'curing' environmental allergies. Please note that immunotherapy has nothing to do with food allergies which can only be managed nutrionally.
The results from the blood panel previously mentioned can be used to create a custom solution containing the most important allergens specific to the dog. The solution is injected or given orally in gradually increasing concentrations. This procedure desensitizes the immune system to said allergens, alleviating the symptoms over time.
It is worth noting that approximately 50% of patients will respond excellent to immunotherapy treatment, 25% will have good response, and 25% will not respond at all. Immunotherapy is no immediate relief and it may take months before any effect is observed. Concurrent treatment is usually necessary for symptom relief. Glucocorticoids should not be administered during immunotherapy as it will considerably decrease the success rate.

This was possibly the most nerve wracking qualifying morning that Eliza had ever faced before. Media day and practices had already been quite overwhelming. Every news outlet wanted to talk to her, about her, and then there were all the brand and team promotions on top of what she was actually there to do. Practices had gone alright, maybe not the best, but they certainly weren’t the worst. Yet Eliza knew she could do better. Maybe it was the nerves or how exhausted she’s been feeling lately, but she needed to shake it off. People were already starting to doubt her and question the team for her joining. She needed to prove herself in this qualifying session and the race.
But first was more time with the media. It was a long morning of getting herself all dolled up. She hadn’t shown up to the paddock wearing anything but something fashionable. She was treating her entrances like a runway, similar to Lewis Hamilton, but she tried to keep it within team colors always wearing something red or with the logo. This time she opted for an all black outfit with some red and gold jewelry pieces. Her hair was straighter and her makeup done lightly, a hint of a pastel papaya colored her eyes.
Once she was done, it was hopping into her car and being driven to the track by a good friend. Alexa happily signed things once she got out of the car and made her way into the paddock, quickly scanning her badge and smiling to all the photographers that snapped her entrance as she made her way to the Ferrari hospitality for her schedule.

2024.05.16 20:58 Hot-Mango-3394 Actor and Model Getting Into Martial Arts

I’m an actor and model turning 25 soon and need to add a martial art to my acting resume. I’m looking for something that combines self-defense, overall fitness, and the ability to compete. Striking arts are preferred in the acting realm but I personally lean towards grappling arts. I understand that for acting, I need to be familiar with the mechanics of strikes, grapples, falls, and impacts to perform fight scenes believably and bring emotional depth to the character.
I’m considering boxing, Muay Thai, BJJ, wrestling, and MMA. I have a reputable and well-established gym nearby that teaches Muay Thai, BJJ, and MMA. Speaking of MMA, they divide it into a wrestling and a striking class. Should I take MMA, given that it includes both striking and grappling, or focus on a specific discipline?
Additionally, I know I can take stage combat classes specifically for acting. However, I’m drawn to the challenge of learning new skills and want something I can compete in as well. What would be the best option to meet these goals? Any advice or personal experiences would be greatly appreciated!

2024.05.16 19:24 FinanceV1global Hiring For Part-Time/Full-Time Executive Assistant

Hey all,
Our firm has tried Indeed, Ziprecruiter, etc but it seems to be a lot of spam resumes so we thought we'd try something a little bit less formal and local!
My partners and I have an accounting firm (taxes) and investment firm. We are a young firm but have been around for over a decade. We specialize in income tax planning, helping clients retire peacefully, and managing investments.
At this point we have grown so fast that we are looking for additional help - we are open to part time and/or full time. Given that we have multiple partners I placed the job title as executive assistant as you would be assisting more than one of us (depending on the season and which department is the busiest).
Job duties include:
-Managing our schedule
-Scheduling clients
-Monitoring email inboxes and taking inbound client calls (assisting them)
-Preparing paperwork (applications, beneficiary changes, investment options, gathering client data)
-Data input in tax software/financial planning software.
-Managing office efficiency (If you notice we can do something bettefaster -- implement it).
-Updating social media and client communications
-Learning new software updates and implementing them
-Scheduling lunch meetings and work events
-Misc tasks that need to be done (going to the post office, mailing out documents, etc.)
The above is just a base sample of what we would need done as an executive assistant; however, if you have an interest in the finance/accounting/world travel field -- there is definitely room to get licensed and learn another skill within our firm... We welcome and appreciate that; so no ceilings would be placed on advancement. This may be learning about stocks & bonds, how to save money on taxes, in's and out's of real estate, life insurance, long term care insurance, Medicare, etc. This would be a very good opportunity for a college student looking to break into the industry - but just to clarify this is a PAID position; not an internship (however it would qualify for college credits - must check with university first).
The ideal candidate is a self-starter and can function independently. We work in a high stress field but our clients are amazing people and we are definitely laid back. If you're open to travel we will pay for you to attend conferences with us (flights, hotels, meals, etc) to help us drive the business forward. We are looking for more of a teammate that is invested in the growth of the firm rather than an employee who just punches the clock. Our goal is to grow together; and see you grow in your knowledge and expertise as well. We take pride in what we do so we would expect the same of you.
Starting pay depends on experience. You would be starting as a 1099 contractor in the beginning to see if this is a good fit for everyone involved. Must have your own transportation and be able to pass a background test and fingerprinting (cannot have felonies/crimes/misdemeanors involving financial matters. We will be implementing a profit share so we can all enjoy success and growth together.
No need for a formal resume as I feel that doesn't tell us much about who you are. A brief summary about yourself, your goals in life, why you'd be interested in working with us, and any questions you may have would be a great start! Please include your name and phone number for a call back.
Our email is: [financialhiringv1@gmail.com](mailto:financialhiringv1@gmail.com)
Thank you Vegas - Talk soon.

2024.05.16 17:18 LastKardax Reflecting on Novelty and Design in TFT Sets: Insights and Thoughts on the Past and Future. (From a nobody's perspective)

Hey everyone! I'm a TFT streamer and Competitive player, hit challenger every set since 4 (not this set! I'll tell you why). I've never posted on Reddit, but I felt like I had too many things I wanted to break down and have a conversation about. These thoughts were just too much for a Twitter post, and I'm really curious to hear how everyone else in the community feels :)!
Disclaimer: I love everyone at the TFT team, and have immense respect for everyone on it, and I know for a fact they all strive to make a product they are proud of. This post is out of love for the game.
  1. Novelty in TFT / Set Mechanics: Regarding Novelty in TFT / Set Mechanics: In recent sets, it's clear that the TFT team is determined to create unique and memorable moments for players, which I appreciate. However, in their pursuit of these moments, they seem to have lost sight of the essence that made TFT so magical from the start.
We can all agree that Set 10 was a huge breath of fresh air coming off the turbo-boring set that was 9.5 where everyone equipped URF legend and popped tome 2-1 and prayed. Set 10 is, in my opinion, one of the greatest sets ever designed. Everything aligned perfectly and harmoniously (music pun!), the unit and trait design was near flawless, the set mechanic was a huge upgrade to Set 4/4.5's Chosen mechanic, and the 5 Costs really did feel like premium units. It was a homerun for TFT. (The numbers show that too for all you numbers people)
Moving on from 10 and coming into 11. It was glaring to me that in an attempt to reach the destination of creating novel moments in TFT they took the wrong road. Encounters might just be one of the worst types of game mechanics to ever exist (second only to shadow items). It's a disaster lottery draw every single time it shows up that you dread won't destroy the path that you calculated was the most optimal path based on what you've gotten so far in terms of units, items, augments, etc... The main reason why Encounters are bad game design is because they take away player agency in an unhealthy way. That doesn't mean that there can't be potential encounter type mechanics that borrow player agency that are healthy. It just means this one is bad. It's a lot of quantity over quality and it leaves the player in this weird middle-ground where they're not too impactful to create a sense of excitement and awe when they pop up, but they're just useful enough to be extremely annoying when they don't benefit you at all, and accelerate other people's boards. (Obviously this isn't ALL encounters, but definitely most)
  1. Unit and Trait design: Set 11 brings some of the most lackluster / boring feeling units and traits I've ever had the un-pleasure of playing. If you named all your favorite TFT units / Traits of all time do you think any of set 11's Units / Traits would make it into your S-Tier category? (Maybe Lissandra, but she's just a worse Set 6 Tahm Kench lets be real) and apart from the idea behind Exalted (It needs more work, but can be an amazing way of introducing infinite comps) not much in Set 11 stands out to me on that front. I'll delve deeper into a few things I don't want to bloat this post, but just know there's a lot more. Kayle is one of, if not the worst summon unit we've ever had in TFT. The idea behind build your own summon unit is fantastic and I genuinely hope they give this another go, but the execution was suboptimal at best. Sets 3 / 4 / 6 / 10 All had extremely life-like and memorable summons Giant Mech Garen that thrusts down sword into the arena, Giant Galio slamming into the arena and punching units to death, Bears and dragons to buff your team and fight alongside you, and even a Hecarim that gallops onto your board and buffs your team and slashes enemies. Summoned units SHOULD feel, sound, and look impactful, and not act like any unit I could buy from the store. Or else they just end up being a glorified 2 cost. Stats aside (I love stats, but this post isn't about them), my primary aim is to ensure a fun game before delving into statistical analysis. Kayle feels like a filler unit that you mostly just plop in the back and forget about, doesn't do much unless you hit the crazy (mostly unobtainable) 10 Piece trait. Speaking of unobtainable traits. It seems to me the TFT team has come up with this bandage solution of gatekeeping a lot of really fun and exciting moments behind these extremely hard-to-obtain 9-10 Piece traits. We've never had this issue in TFT; why are we now gatekeeping a lot of the exciting moments behind 100+ hours of playtime. You can tone down a lot of the exciting moments, and still keep them accessible, but challenging to obtain. You've done this for the longest time; why fix something that isn't broken. Personally, I think there needs to be a revaluation of what makes a Set engaging and fun, and what players really want to see and play around with for hundreds of hours way before you start delving into the mindset of "How do we add as much divergence, nuance, and novelty". Those roads can meet and intertwine, but they aren't the same road.
  2. Items, Augments, and Orbs!: Items - Here's a little brain exercise for everyone, what would happen in TFT if we removed all item components, and strictly dropped only Ornn anvils all game? (Take a minute to think before you click the spoiler!) If you guessed it would ensue chaos, bugs, and a balancing nightmare. OR be a hell of a lot of fun! you'd be correct both ways! (New portal idea!/s) Luckily for us, that isn't the case. However, I do think we've reached a point with items where we're too comfortable with what they wish to achieve on units, and this creates a dynamic where TFT units are always being designed with items in mind, and how the items would make the unit work. Rather than creating fun and engaging units in a vacuum and worrying about the items later. It's a scary prospect to experiment with the tried and true (Think back to Set 7 when they made Rage-wing trait experimenting with units that had a different fuel source than mana, but it was a total miss or when they created the dreaded Shadow items), but it's just food for thought. Just because you didn't get it right once, doesn't mean the idea is flawed, just means you didn't get it right that one time. A time where you definitely did get it right however, is Radiant items Look how those turned out! Some of the most beloved items of all time. This is sort of what they wanted to achieve with the new expansion to the Ornn anvil items, and support items. It's definitely a step in the right direction, but can definitely be explored more.
Augments - Augments really was a happy little accidental shift in TFT's life cycle. They created the perfect concoction of how to add an almost infinite amount of variability to a game that was missing just that. (Which also adds to why Encounters are redundant and frustrating) Since then they've time and time again created really fun and engaging augments each set, that without I'm almost certain TFT wouldn't be as popular as it is. Please keep doing this, this is really good. Another little thought exercise before we continue. What would happen if most of the Encounters were just turned into Augments instead? (Think a little before you click spoiler!) If You guessed it would just create a healthier way of interacting with said encounters. You'd be correct!
If you're trying to create a new set mechanic (encounters) it shouldn't overlap with something that already exists that's just better than the new thing you're trying to create (augments). However, getting back on track. It does feel like things have gotten a little too comfortable in the Augment space, and feels like there hasn't been much room for innovative change. There's an untapped reservoir right in front of you of infinite variability at your disposal. Why aren't you experimenting harder, instead of looking for other means of adding variability that will always be suboptimal to the one you've already created almost 6 Sets ago...?
Soft Sample Examples: A new type of Augment called Radiant Augments that provide a wide range of very strong / borderline insane combinations of items and stats, but the catch is after every player combat if you lose you can choose if you want to swap your Radiant augment with the player you just faced. (Like multiple random hexes of strong stats, or 3 tailored Orrn items / Support items, or given 3 zephyrs and knowing who you're fighting next or given a 2Star-dragon that adopts your top-most trait?!) (The design space is infinite)
Orbs - Lastly, I'd like to talk about Orbs! Orbs are a lot better, good job! That's all I have to say :)!
Thank you for reading this far into my post if you have. Again, this comes from an immense love for the game and the people creating it. I have nothing but positive interactions with Mortdog on Twitter / Discord and would appreciate if you guys keep it civil and friendly :)! I really do have a lot more thoughts, and things I'd love to share, but I feel like I've already bloated this post a lot! (Assassins, Higher Cost units than 5, Midsets, system changes, loot management, meta knowledge, Portals etc..)
I've just been really bored playing this set, and that's the reason I've retired from seriously competing or climbing for this one set, and I'm very keen to read what other people have to say about my thoughts, and looking forward to discussing it thank you!

2024.05.16 16:50 DarkEsca Time Machine to RBY #3: Cinccino

Time Machine to RBY #3: Cinccino
image thumbnails are still a pain
This awesome RBY-style Cinccino sprite is made by SharkGuy01--check them out on DeviantArt!
In this series we will analyze how modern Pokémon would function if they existed back in RBY. Note that I will only cover efficiency in RBY OU specifically, as I don't have enough knowledge of RBY lower tier metas to say much with confidence about them. As for the rules when "translating" a mon to RBY, check out the first installment.
For the third entry of the series we will take a step back from tossing high-BST legendaries into the good ol' days and throw in a more innocent little critter. A rather unimpressive mon outside of the kawaii factor, Cinccino doesn't exactly scary many people in modern OU metagames, and when transporting it back to RBY it would lose many important moves like Tidy Up, Triple Axel, Rock Blast and even its signature Tail Slap. Will the coveted Normal type and some hidden gems in that movepool be enough to make it usable? Let's find out.

The Stats

Jumping out is the 115 base speed. This is excellent by RBY standards, outpacing Tauros and speed tying with Starmie. Alakazam and Jolteon are the only relevant things that threaten to outrun you and neither are known to be good at tanking physical hits. You also get a crit rate of roughly 22% which is nice to have.
95 Attack is solidly usable. It's not terribly high, but remember Tauros dominates the meta with only base 100. With STAB Hyper Beam under the belt, rest assured you can threaten some things.
The good parts end there though, as its bulk is... really not good. 75/60 physical bulk is barely better than Jolteon, and low enough that Body Slam into Hyper Beam from Tauros is a KO more often than not, pretty much invalidating your status as a potential switchin to it despite immunity to Body Slam paralysis. Rhydon Earthquake also always 2HKOs, and you often don't even take an Exeggutor Explosion from full HP. It's not as horrid as it is in later gens due to RBY bulk mechanics, but it's definitely far below what the rest of RBY has.
65 Special is also definitely not something to write home about. Its special attacks won't threaten many things unless they're x4 effective or something, and its special bulk isn't too great either. Alakazam's Psychic is a roll to 2HKO without any crits, for example. Granted, this special bulk is comparable to Tauros', so it's not like this is unheard-of levels of low, but it's a definite downside and quite the uphill battle to overcome.

The Typing

There's not much to say here. Normal has a case for best type in RBY. You lack exploitable weaknesses due to Fighting moves being nearly inexistent, so despite being on the frail side, you're almost never going to be OHKOd outright. In addition, you cannot be paralyzed by opposing Body Slam. This greatly improves your matchup into opposing Normals, as while you still take more actual damage from Body Slam than you'd like, you cannot be trolled by paralysis in a 1v1. Offensively, Normal very notably gives you STAB on Hyper Beam, meaning you have a powerful finisher that can threaten moderately chipped stuff to switch out. It also would give you STAB on Body Slam (the 'would' is an omen to the unfortunateness coming later).

The Movepool

Looking at the standard offensive movepool, we notice a big problem: Cinccino does not get Body Slam! This is a huge blow to its general offensive capabilities: in cases where Hyper Beam doesn't kill yet, you are now reliant on Double-Edge for damage, which is admittedly stronger but also has very annoying recoil damage that only exacerbates your subpar bulk. In addition, Double-Edge cannot threaten to paralyze switchins like Starmie and Rhydon, which complicates getting past them in the long run. You do have Hyper Beam, at least.
Your coverage is also pretty pathetic. Only having Thunderbolt and Thunder, coming off 65 Special, is poor. The only real target for these is Cloyster, which you threaten a 2HKO on with Thunder. This is pretty notable since Cloyster otherwise looks like it walls you fairly hard (especially since you cannot even threaten to paralyze it). Other potential targets will be less impressed, like Starmie, who is only 3HKOd by both Thunder and TBolt.
That being said, our little chinchilla rat has three notable moves not mentioned yet that might well elevate it beyond "shitty Tauros with 5 more speed".
The first of these three is TWave. TWave somewhat alleviates the lack of Body Slam paralysis by letting Cinccino threaten to just manually paralyze stuff. Being rather frail and running into 4MSS as we'll see later makes this not the hugest deal, but it's still a very handy tool to have access to and pull out once in a while.
The second hidden gem is Sing. Sing immediately gives Cinccino a massive niche: since it outpaces Gengar, this makes Cinccino the fastest user of a sleep move in RBY. Of course Sing isn't exactly accurate, but the threat of it alone will influence play a lot, and this would definitely give Cinc a spot on at least some of those Gengar Lead teams. It's unlikely to push Gengar out of the meta altogether, since it lacks that notable Normal immunity that Gengar contributes to teams, but people looking for high-risk-high-reward strategies that want the fastest Sleeper out there now have a new option.
The third, and perhaps most notable move, is Super Fang. Super Fang in RBY is... really cracked.
To put things into perspective, Super Fang was enough to put Raticate on the OU VR for a long time. Because everything in RBY is absurdly bulky, even the meta's strongest attackers often only 3HKO their targets. What prevents Tauros from just running through everything on turn 1 is that most things do in fact take Body Slam into Hyper Beam from full HP if no crits are involved. In an environment like this, a move that just cuts the opponent's HP in half is massive. Cinccino is barely strong enough that its Hyper Beams generally do around 50% to most neutral targets that aren't massively bulky, including Starmie, Tauros, Chansey, Zapdos, and Exeggutor. I think it becomes pretty clear what we're going for here. There are very, very little things that can safely switch into Super Fang and not be at risk of dying to a followup Hyper Beam. On a mon that outpaces Tauros and isn't Raticate levels of frail, this is... pretty fucking huge.
Super Fang pushes Cinccino from being down with mons like Dodrio and Kangaskhan as "I wish I was Tauros" to a genuinely huge offensive threat in its own right. It's pretty risky to use because of Super Fang accuracy and needing to get predictions between Super Fang and Hyper Beam right a lot, and truthfully most of the mons mentioned aren't guaranteed to die to Super Fang->Hyper Beam from full HP (unless a crit bails you out) but with a bit of luck the chinchilla will definitely tear massive holes into the opposition. And if you thought it was bad enough as a Tauros replacement, imagine it as a Tauros partner. Even if Cinccino dies in a 1v1 it could never win, the bull in the back will gladly take on the Rhydon or Snorlax it forced to low HP and do Tauros things afterwards.

Checks & Counters

The biggest counter to Cinccino is without a doubt Rhydon. Super Fang will massively chip it which is not appreciated in the slightest if Rhydon wants to deal with Cinc's teammates or get Substitutes up later, but beyond that, Cinc isn't doing much to it. A movepool consisting entirely of Normal and Electric moves can obviously not touch Rhydon for relevant damage, with a crit Hyper Beam capping at 38% and a non-crit one not even KOing after two Super Fangs.
Gengar provides similar issues. Interestingly, Ghost isn't immune to Super Fang in RBY, so it still takes hefty damage from it, but Cinc's moves that actually finish a mon off after Super Fang can't really touch it. The most it can do is Thunder, which without a lucky crit caps out at 20%. Gengar meanwhile has a good shot (factoring in Gengar's crit rate) to 3HKO back with Thunderbolt or Psychic of its own or can just Explode for roughly 75% right away.
Very physically bulky things like Lapras, Cloyster (if no Thunder) and Snorlax can win the 1v1 by tanking Super Fang into non-crit Hyper Beam, if they have options to 2HKO Cinccino back. They can't exactly hard switch in though, since a second Super Fang (or just a Double-Edge) will put them in range of Hyper Beam afterwards. This also means stalling Cinccino out with Rest isn't exactly viable, since Super Fang->Super Fang->Hyper Beam will just KO.
Alakazam and Jolteon can serve as checks by virtue of outrunning Cinccino and 2HKOing (in Alakazam's case) or threatening to paralyze, which immediately makes Cinccino's threat level plummet by making it unable to overcome foes with recovery without paralyzing them itself. Needless to say though, neither of them has any business hard switching in: Jolteon will always fall to Super Fang -> Hyper Beam even from full HP (with the tiniest bit of chip, even Double-Edge does the job) and while Alakazam can play mindgames with Recover and potentially Reflect, Cinccino's high crit rate and Alakazam taking at least 80% from even non-crit Hyper Beam turns this into a game it really wouldn't want to play.
While not outrunning it and not at all being a safe answer, Starmie also annoys Cinccino because of the speed tie making a lot of its options unsafe to go for. If it Hyper Beams on a turn that Starmie outruns and Recovers, this gives Starmie a free turn to do whatever it wants, often paralyzing it. Clicking Super Fang on a Starmie that's slower that turn on the other hand wastes Super Fang PP and Starmie will still be out of Hyper Beam range on the following turn. If Cinc spends a turn paralyzing Starmie with Thunder Wave, chances are Starmie will paralyze it back the first chance it gets. And in a raw 1v1 where both mons got a safe entry, they're likely exchanging TWaves as well, which Cinccino hates way more than Starmie does.
Lastly, rather than looking for hardcounters, a good way of dealing with Cinccino would probably be to just outplay and outrpredict it. Super Fang into Hyper Beam gets many KOs, but Hyper Beam on its own gets little. This puts Cinccino in prediction-heavy situations a lot since not getting a KO with Hyper Beam means you're taking a heavy punish, but Super Fanging something that stays in will also have uneasy consequences. You're not Raticate levels of frail, but still take heavy enough damage from many things that you want to be careful with your HP, and if something TWaves you it's as good as game over usually. Couple this with Cinccino not being able to hard switch into most things safely, and Super Fang missing from time to time, and it makes these scary interactions perhaps a bit less likely to happen in practice.

Conclusion

Raticate, whenever it's ranked, usually lingers down in E because even Super Fang has trouble truly popping off on a mon with stats that atrocious. However, we've seen that with Cinccino, which is way faster and just strong enough to pick up many 2HKOs that Raticate lacks, the genuine threat level is there. The mon is pretty cheesey still because of a reliance on good prediction and luck, but teams without a Rhydon are definitely in trouble against a well-played Cinc, and if nothing else the holes it opens up for a teammate like Tauros in the back are easy to exploit.
It's tough to imagine Cinccino being actually reliable and splashable enough to be on par with the A Ranks, but the threat level is likely enough to hang in the lower echelons of B1 still around Cloyster and Jynx's level.
As for how the rest of the meta might adapt to Cinccino's presence, Rhydon might see an increase in usage as probably the hardest answer to it, though being tasked with answering Cinccino does greatly cut into its capability to deal with Chansey later (funnily a Cinccino could kamikaze into an incoming Rhydon with Super Fang -> Super Fang -> Hyper Beam to put it into Zapdos Drill Peck range though, but this isn't likely to be worth it). Gengar is a mixed bag: on one hand it also beats Cinccino decently well, but on the other it faces competition from it as a fast sleeper, so its total usage might not change too much. Snorlax might run Hyper Beam a bit more so that it can properly 2HKO Cinccino with Body Slam -> Hyper Beam (or just Hyper Beam an already chipped one). Lastly a lot of "second Tauros" mons in the meta will likely greatly fall off if not become irrelevant altogether. Kangaskhan has a bit of a case to stay with its good bulk and Rhydon matchup, but Persian has it rougher since it lost its speed tier advantage and Super Fang does more than Slash on average. Raticate, unsurprisingly, would never even be considered for the E Ranks again, its minuscule niche of hitting Rhydon with Bubble Beam not nearly being enough to make up for everything it loses compared to Cinc.
What do you think? Would scarfy have an easier or harder time doing stuff in RBY than I imagined? Thanks for reading!

2024.05.16 16:34 pretty-in-pink Possibly at a crossroads with a career option. How can I leverage it for actual work I want to do in the future?

I (26F) have been having employment issues for over a year despite having a masters degree in public administration/certificate in health administration and top grades (long story short I had two job that I was not qualified for and they hired me anyways, only for them to fire me afterwards a few months later leading to a gap).
I got to the point that I was applying to ANY job just to get the skills/references. And then I sent a general application to a religious elementary school I used to go to.
I had an initial call with them and it all went well, I am going for a trial day next Thursday to see if I am the right fit . There were no administrative positions but there were open spots for assistant teachers in the Early Childhood Department for Pre-K.
Now this didn't bother me because:
  1. I am close friends with someone who works in the department
  2. My capstone was about children's education during distance learning and future economic prospects
  3. I have extensive volunteer experience working in my local temples children's program and very good references
  4. Salary (which can be $19 according to the school's Glassdoor) is not my biggest concern at the moment, my concern is getting work/references.
5)Schedule is flexible aside from departmental meetings
6) Doesn't require me to get any teaching degrees, only certifications in the state (NY) that I'm currently in.
However I am worried about the future as well because I hope to eventually move to Washington DC in a couple years, which will mean I have to leave the job eventually and find a new one. And I'm worried by taking this position as a Pre-K assistant teacher I'm nuking any chances of ever working directly within the policy world or for local communities.
My questions really are then:
  1. Should I take the job if offered? Most likely I won't be starting until the 2024-2025 semester which starts in September. Which gives me three months not looking for a job in anticipation of working there. I live with my parents so rent and food is not an issue for now but it'll be a hard stop on spending. If so....
  2. Should I take the job if offered but let them know if I get a position in my field by the summer more relevant I have to decline it?
  3. If I take the job, how can I leverage it for work I want to do, especially since its an assistant teacher job for Pre-K? Just searching around I could get involved in my local religious community more when it comes to work but they also require office experience. What about writing samples? How could I realistically leverage this position for other teaching jobs with better pay in the future?Another option could be local community centers?
My main issue regarding it all is that I mainly have internship experience currently on my resume for the more administrative jobs which may tank other prospects in the future for me.

2024.05.16 16:10 RedChipCompanies BullFrog AI and Lieber Institute for Brain Development Collaboration Identifies Novel Drug Targets for Neuropsychiatric Disorders

GAITHERSBURG, Md., May 16, 2024 (GLOBE NEWSWIRE) -- BullFrog AI Holdings, Inc. (NASDAQ: BFRG; BFRGW) ("BullFrog AI" or the "Company"), a technology-enabled drug discovery company using artificial intelligence (AI) and machine learning to enable the successful discovery and development of pharmaceuticals and biologics, today announced significant advancements in its collaboration with the Lieber Institute for Brain Development (LIBD), including the identification of potential drug targets for multiple neuropsychiatric conditions.
BullFrog AI and LIBD have made remarkable progress in identifying novel subgroups between and within neuropsychiatric disorders, including major depression, schizophrenia, and bipolar disorder. Utilizing a combination of ensemble machine learning, generative AI, and graph analytics, the team has successfully clustered patients by expression levels of gene isoforms across multiple brain regions. This innovative approach led to the identification of dozens of clusters of patients, each exhibiting differential enrichment of neuropsychiatric conditions, providing unprecedented insights into the biology of these disorders. These biological subtypes could ultimately lead to targeted therapeutics for a more precise treatment of psychiatric disorders.
For each identified cluster, key genes have been pinpointed that explain cluster membership. These genes present new potential drug targets by revealing unique molecular mechanisms and pathways associated with each cluster. To prioritize these genes for wet lab validation, BullFrog AI is now applying Causal AI, a crucial step for confirming the therapeutic potential of the identified targets.
“This collaboration continues to yield transformative insights into the biological underpinnings of neuropsychiatric disorders,” said Vin Singh, CEO of BullFrog AI. “The identification of these novel subgroups and key genes is a testament to the power of AI in advancing precision medicine. Importantly, our proprietary bfLEAP™ platform, combined with LIBD’s unparalleled brain data, is paving the way for the development of targeted and effective treatments, and we have initiated engagement with pharmaceutical companies with an objective of securing multiple strategic partnerships in the coming quarters.”
Daniel R. Weinberger, M.D., Director and CEO of LIBD, added, “Our partnership with BullFrog AI has unlocked new pathways for understanding the complexities of brain disorders. These findings not only advance our scientific knowledge but also open new avenues for developing targeted therapies that can improve patient outcomes. We are excited about the potential impact this collaboration holds for the future of neuropsychiatric treatments.”
The collaboration between BullFrog AI and LIBD, announced in September 2023, leverages the bfLEAP™ platform to mine LIBD’s comprehensive brain data. This data includes transcriptomic, genomic, DNA methylation, cell-line, clinical, and imaging data from over 2,800 brain samples. Early results announced in January 2024 highlighted the ability to stratify brain expression data, revealing biological subtypes within psychiatric disorders.
The potential for new treatments for psychiatric disorders is vast and underserved. With increasing global awareness and demand for mental health solutions, this breakthrough presents a potential paradigm shift in treatment approaches. By identifying possible biological subtypes within disorders, BullFrog AI and LIBD are not only advancing scientific understanding, but also opening doors to novel therapeutic pathways and personalized treatment strategies.
The three-year agreement between the organizations granted BullFrog AI a period of exclusive access to LIBD’s brain data with the potential to commercialize products or services derived from the collaboration. The partnership represents a unique synergy between AI-driven analyses and world-class neuropsychiatric research data, setting a new standard in the pursuit of effective treatments for brain disorders.

2024.05.16 14:59 Sea-Steak-4422 frustrated gamer playing ac origins

Hi Guys. l would appreciate any feedback from people who have played ‘origins’ as l’m finding it all very frustrating to the point of stopping. My jibes are the following: My level is 15 and having to ‘kill all the looters’ in a task has been daunting and frustrating. l can’t seem to pass it so for example most of the ‘looters’ are either my level o one level above. l can fire an aimless amount of arrows at a level 16 and the guy is still walking around with tons of arrows in his back it takes an eternity to finish him. l then have to ‘fast travel’ to ‘refill’ my arrows then proceed back for the next one. if there are say 8 Bandits and l kill 6 with great effort then ‘die’ my game then reverts back to the 8 bandits and l need to start again. so all the bandits that were ‘ dead’ lying around, have all resumed their positions again. l can give attack after attack either with sword o arrows to kill one but should they hit me l’m dead after 2 hits. l’ve played ‘skyrim’ and its so much better to kill and ‘quicksave’ furthermore you collect ‘gear and arrows’ all the time. what also ‘ bugs me’ is l have gained alot of ‘items’ through the quests so l can sell items no problem but if l go into my ‘gear to select a better sword o bow they are not there to use. Am l doing something wrong how can l have all these better items but still have to use the bronze sword and basic bow which l started with. l also find the ‘mechanics’ of the game a strain skyrim although in my opinion was abit ‘boring’ the mechanics of selecting and using gear was so much better and the ‘quicksave’ was really good. finally, am l right in thinking to ‘upgrade’ gear you have to run around killing animals for pelts etc? this is crazy in my opinion. l played earlier ac games and enjoyed them but this game for me sucks. playing on a ps5 thanks

2024.05.16 14:55 ant_queeen How to break into field with basic lab ecology/evolution background?

I just finished my masters in ecology/evolution a year ago and have been working as an underpaid research assistant in the biomedical sciences. It’s been super depressing/demotivating and after a lot of thought I am trying to break into the field of environmental science- ideally a position in research where I can use my lab or writing skills. I tailor my resume to each position but I am having trouble landing an interview. I am even applying for consulting and air pollution jobs with no luck- probably more likely to hire ppl with environmental science degrees. I completed a cQHEI stream assessment course but can’t really afford the wetland delineation course as it’s $500+. I am not wanting to move and desperately need a career change for my mental health and career trajectory. Any tips on how to find a job?
Edit: I also have some field sampling experience with tropical insects and soil, a lot of teaching experience, and am developing knowledge of wetland/native plants and animals in my spare time.
Edit2: I’m not sure why my comments are being downvoted. I’m doing all I can to have a competitive resume and find a job within my constraints. If your opinion is that the only way to get a job in this field is to move and take a bunch of expensive courses then please just say so!

2024.05.16 14:25 sebastiandang How License and Capacity Unit works?

Hi folks, after reading a lot docs about the `License` and `Capacity Unit` I'm still not understanding much about the mechanism and how it charging my CUs? Sample: With the lowest Supcription -> 2CUs -> Which means? 2CUS *(2592000s in a month) -> so I have ~ 5Mils CUs. If this is correct, here is my situation: (1) I have notebook that not work correctly and its consumes a lot CUs, which makes all the operations cannot work *exceeded CU. But after checking out the Report about CUs - this month Im using only 1.5Ms of CUs. I was though it the total / 30days, so it might be recover the day after, but its not! (2) If I start a pipeline, other notebooks cannot start a Session also - even though it was configured with 2 nodes only! I have read the the "overhead" mechanism. But not understanding that much! Thanks for reading this!

###

It includes that last line in the output, so it might be fun to scan your FrankenSPAM for tell-tale filler text at the end like that.
What have we learned? AI-text is basically easy to spot, and is really not that different from the years of fill-in-the-blank templates and business mail that commerce has deployed for decades. It sounds canned and trite, because it's the "elevator music" of text.
I have seen a lot of work posts on UpWork just like this, and indeed on dozens of other job-board sites. I love the emphasis on "story-teller" and "narrative," which do ring true as the hot content-creator buzzwords of 2024.

2024.05.16 12:26 Microlit_ What measures are taken for quality control in the usage of micropipettes to ensure accuracy and reproducibility?

About Micropipettes
Micropipettes are one of the most significant tools in a laboratory setting that facilitate precise and accurate measurement of liquids in microliter volumes. Whether you are dispensing reagents for experiments or preparing samples for analysis, the reliability of micropipettes is of high importance in ensuring the integrity of scientific results.
To maintain accuracy and precision in pipetting operations, stringent quality control measures are implemented across laboratories. Here are some of the measures that can guide you through the quality control & usage of micropipettes.
Calibration Process:
Calibration is the base for micropipette quality control that helps to ensure pipette calibration standards and deliver accurate volumes as specified in the laboratory. In the same manner, regular calibration is essential to account for factors such as wear and tear, environmental conditions, and operator technique.
Pipettes calibration is done using gravimetric or volumetric methods, where the dispensed volume is compared against a known standard. By calibrating pipettes at specified intervals, laboratories can minimize measurement errors and maintain consistency in pipetting accuracy.
Routine Maintenance:
Routine maintenance of micropipette technology is crucial for prolonging their lifespan and preserving accuracy. This includes cleaning, lubrication, and inspection of critical components such as seals, pistons, and O-rings.
Regular maintenance tasks prevent contamination, ensure smooth operation, and detect any signs of wear or damage. Additionally, calibration verification checks are performed during maintenance to confirm that pipettes remain within acceptable tolerance limits.
Operator Training & Proficiency:
Effective training and proficiency testing are essential components of micropipette quality control programs. Proper pipetting technique, including pipette handling, sample aspiration, and dispensing, is taught to laboratory personnel to minimize errors and variability.
Proficiency assessments, such as pipetting accuracy tests and precision exercises, validate operator competency and ensure adherence to standard operating procedures (SOPs). Ongoing training and refresher courses are provided to reinforce good pipetting practices and address any performance deficiencies.
Environmental Conditions:
Environmental conditions can significantly impact micropipette accuracy and reliability. Factors such as temperature, humidity, and air pressure can affect the viscosity and surface tension of liquids, leading to volume discrepancies and mechanics of micropipette.
To mitigate these effects, laboratories maintain controlled environments within specified tolerance ranges. Temperature-controlled pipette calibration rooms and humidity monitoring systems help maintain optimal conditions for pipetting operations, minimizing the risk of measurement errors due to environmental fluctuations.
Usage Guidelines and SOPs:
Establishing comprehensive usage guidelines and SOPs is essential for standardizing pipetting procedures and ensuring consistency across laboratory workflows. SOPs outline proper pipette usage, maintenance protocols, calibration procedures, and troubleshooting guidelines.
By adhering to SOPs, laboratory personnel minimize variability in pipetting practices and promote reliability in experimental outcomes. Regular review and updates to SOPs incorporate best practices and lessons learned, further enhancing quality control measures for micropipette uses.
Data Documentation & Traceability:
Accurate documentation and traceability of pipetting activities are critical for quality control and regulatory compliance. Laboratories maintain detailed records of pipette calibrations, maintenance activities, operator training, and pipetting protocols.
Electronic pipette management systems track pipette usage history, calibration schedules, and performance trends, ensuring traceability of measurement data and facilitating audits or inspections.
Conclusion:
Talking about the quality control in the usage of micropipettes, know that it’s essential for maintaining accuracy and reliability in laboratory operations. Thus, by implementing rigorous calibration procedures, routine maintenance protocols, operator training initiatives, environmental controls, usage guidelines, and robust data documentation practices, laboratories ensure the reliability of pipetting results. These quality control measures uphold the integrity of scientific research, promote confidence in experimental data, and contribute to advancements in various fields of science and technology.

2024.05.16 12:02 tung0310 Late-Night Eating and Its Impact on Obesity

Obesity affects about 42% of the adult population in the United States and contributes to the onset of chronic diseases such as diabetes, cancer, and other conditions. Understanding the mechanisms behind this is crucial to combating the obesity epidemic. "We wanted to explore the mechanisms that might explain why late eating increases the risk of obesity," said Dr. Frank A. J. L. Scheer, the senior author of the study. Previous research indicated that late eating is associated with increased body fat and less successful weight loss.
The Study: Timing Matters The researchers asked, "Does the timing of our meals matter when all other factors are consistent?" The answer was a clear yes. Eating just four hours later than usual significantly affected hunger levels, how calories are burned after eating, and how the body stores fat.
In their controlled lab study, the team worked with 16 participants with a Body Mass Index (BMI) in the overweight or obese range. Each participant followed two eating schedules in the lab:
Normal Schedule: Eating at typical meal times. Late Schedule: Similar meals but eaten 4 hours later than usual. Before the lab sessions, participants maintained a fixed sleep-wake schedule for two to three weeks, and for the final three days, they followed a strict diet to standardize intake.
Measurements and Observations In the lab, participants reported their feelings of hunger and cravings, provided frequent small blood samples throughout the day, and had their body temperature and energy expenditure measured. To understand how meal timing affects fat storage pathways, the researchers also took fat tissue biopsies from a subset of participants under both early and late eating conditions for genetic expression analysis.
Study Findings: Hormones, Calories, and Fat Storage The study revealed profound effects of late eating on hunger and the appetite hormones leptin and ghrelin:
Leptin Levels: This satiety hormone was lower over 24 hours during the late eating condition compared to early eating. Calorie Burning: Participants burned calories at a slower rate when following the late eating schedule. Fat Storage Genes: Gene expression in fat tissue showed increased fat creation and decreased breakdown (lipolysis) under late eating conditions. These findings highlight the physiological and molecular mechanisms underlying the correlation between late eating and increased obesity risk.
Implications and Insights These findings align with a substantial body of research indicating that late-night eating can increase a person's likelihood of developing obesity. They elucidate how this might happen by revealing changes in different control systems related to energy balance. By conducting a randomized crossover study and tightly controlling behavioral and environmental factors like physical activity, posture, sleep, and light exposure, the investigators could pinpoint changes in different systems that control energy balance, providing a clearer picture of how our bodies use the food we consume.
This research suggests that modifying our eating times could be a straightforward approach to reduce obesity risk, improve metabolic health, and better manage appetite and fat storage.
Conclusion The study emphasizes that not just what we eat, but when we eat, plays a significant role in health outcomes related to obesity. Shifting meal times earlier could be a viable strategy for reducing hunger, improving metabolic rate, and ultimately aiding in weight management and obesity prevention.

2024.05.16 11:01 ShityTrashCan What species could Ben Tennyson turn into?

Which of the following species could Ben turn into & couldn’t turn into? I know there’s the Ink Tank video that explains things but I’m still iffy about some & can’t really make answer.
Rules for the Omnimatrix : 1)The being in question must be sapient 2)The being in question must have DNA or an equivalent 3)The being in question must be different enough from an already existing transformation
Species : - Imaginary Friend (Fosters Home for Imaginary Friends) - Watermelon Steven & Animals (Steven Universe) - Pumpkin (Steven Universe) - Shin Godzilla (Shin Godzilla) - Life Fibers (Kill La Kill) - Pillar Men (JOJO) - Ted (Ted) - Stitch & all the other Experiments (Disney) - Titans (Owl House) - Palisman (Owl House) - Ponyo (Ponyo) - Eva Units (Evangelion) - Gru’s Dog (Despicable Me) - Salmonid, more specifically King Salmonids (Splatoon) - Brain (Pinky & the Brain) - Penguins (Penguins of Madagascar) - Perry the Platypus (Phineas & Ferb) - Winston (Overwatch) - Hammond (Overwatch)
Notable things for some species : - Imaginary friends are created through the imagination of a child who desires a friend. Imaginary friends can come in all shapes, sizes, species, etc. and their creation and form don't seem to be limited by anything so long as the child can think it, it can happen. They can be an anthropomorphic object, including a food or something from a fantasy book. - Watermelon Stevens are a species of sentient watermelons accidentally created by Steven. They are fully-formed sentient lifeforms that are capable of feeling emotion and gestural communication. Genetic derivatives like watermelon chickens and dogs exist, presumably from the Watermelon Stevens. - Shin-Godzilla originated as a prehistoric marine animal of some kind, proposed by Professor Goro Maki to be a relative of Paleozoic marine reptiles, which managed to survive into the 20th century. When the United States dumped barrels of nuclear waste into Tokyo Bay in the 1950s, the creature began feeding on this waste, causing him to mutate. - Life Fibers are multi-cellular microorganisms. Life Fibers are parasitic organisms, living off the bio-energy that exists within the cells of the nervous system as well as in the circulatory system. Life Fibers also seem to be self-aware entities, capable of making decisions and thinking. Life Fibers reproduce by influencing a species evolution to become the dominant species. After the species has spread across its planets, it’ll take over the hosts body & rise into the stratosphere to create a massive cocoon of Life Fibers that envelops the entire planet. - Jumba reuses DNA from other experiments as bases/prototypes for other experiments. Each genetic experiments was created with multiple kinds of DNA that Jumba sampled throughout the galaxy so they all age differently. - Palisman are a sentient magical totem that sits at the top of a magic staff. A palisman contains a magical essence and harbors various magical properties, including the ability to switch between wooden and living forms at will. - All Evangelion Units are living creatures that have a number of electronic and mechanical modifications done to them to essentially keeps them from going berserk and allows their pilot to control them. - Kyle (Gru’s dog) is not a dog. The scanner of Anti-Villain League can't recognize which species he belongs to, and his age far exceeds that of regular dogs. - Winston & Hammond are genetically altered and super-intelligent animals that were sent to live in a colony on the moon. - The Omnitrix can scan & transform into lab rats & a dolphins.
